Default Wrap Text

When I format certain cells with wrap text, even though the data fits on one
line, the first line is blank and the data is on the second line. Why is
that?

Could it be just your vertical alignment setting?

Default Wrap Text

Example:
I open an empty Excel file. In A1 I type,
Core System Control: Facility Object Manage,
which all fits. The width of column A is 38.76, and the height of row 1 is
12.75. The Font used is Arial Regular 10. When I format A1 and set wrap
text, a blank line appears above the text.

Can you put the cursor in that blank line ?
Still seems like it's just your alignment setting being "bottom" and the row
being taller than needed.

If you double-click on the lower margin of the row number on the left it
should autofit and the space should disappear.

I don't know what you mean by "the row being taller than needed." The row
size was fine until I set the wrap text flag. I tried "double-click on the
lower margin of the row number on the left it", but I saw no change. Also, I
am setting the wrap text with a macro, which sets the whole column.

I have had the same problem. Something that seems to fix it, at least
temporarily, is to de-select wrap text, resize the column width to autofit,
then select the column, re-select wrap text, and then do the row autofit
again.
