Default How do I compare two lists of numbers?

I have a long list of numbers in two columns. I want to find out what
numbers are in column A that are not in column B. Is it possible to use
conditional formatting to do that?

Yes, Select column A, and use a formula of

=COUNTIF($B:$B,A1)=0
