The phrase "significantly different" usually applies to statistical
inference. Consult a book. You'll need to make assumptions about the
underlying population distributions, and you'll need to know the sample
sizes of the random samples.
If you want Excel to do the calculations for you, one way is to start with
the data for the two random samples in columns on a worksheet, choose Tools
| Data Analysis, and pick the appropriate t-test.
