Default Can you enable auto complete from a custom list in Excel?

No.

Autocomplete or "Pick from list" gets its information from cells above and
below the point in the same column that you are entering new data.

A custom list entered in ToolsOptionsCustom Lists is used for autofilling
and when sorting.
